Evaluation of antidiabetic potential of combined extract of Morinda citrifolia L fruit and Annona squamosa L leaf in alloxan induced diabetic rats

Abstract: This study investigated the effects of Metformin, McF+ASqL Mixed Extract (which contains active compounds like alkaloids and flavonoids), and their combination on diabetic rats over 28 days. Five groups were tested: normal control, diabetic control, Metformin-treated, McF+ASqL treated, and combination-treated. The diabetic control group had higher blood glucose levels. Metformin, McF+ASqL, and the combination treatment all reduced glucose levels, with the combination showing the most significant reduction, bringing glucose levels close to those of the normal control group by day 28. Significant improvements were also observed in the lipid profile of treated groups. Groups III, IV, and V showed reductions in serum LDL and increases in HDL, along with reduced triglyceride and cholesterol levels. Group V exhibited the most pronounced effects, although all treated groups still had lipid levels higher than normal. Additionally, liver function markers, such as ALT and AST, were elevated in Group II. Treatment with Metformin, McF+ASqL Mixed Extract, and their combination resulted in reductions in these enzymes, with Group V showing the most notable improvement, though levels remained above normal. Hemoglobin and packed cell volume (PCV) levels were significantly lower in Group II compared to Group I, but treatment with Metformin, McF+ASqL Composite Extract, and their combination led to improvements, with Group V achieving normal hemoglobin and PCV levels. Liver glycogen levels were also significantly improved in all treated groups compared to Group II, with the combination treatment showing the most significant increase, though still slightly below normal level. Additionally, the combination treatment (Group V) showed the most significant improvement in total protein levels, and reductions in BUN and serum creatinine, although BUN levels remained elevated in all treated groups. Overall, the combination of Metformin and McF+ASqL Combined Extract demonstrated superior therapeutic effects in improving multiple physiological parameters affected by diabetes. Histopathological findings demonstrate that the combined treatment of AsqL, McF, and metformin alleviated degenerative changes in the liver, pancreas, and kidneys of diabetic rats, suggesting potential therapeutic benefits.
