Acute oral toxicity study of thiamine rich herbosome in rats

Abstract: Vitamin B1 or Thiamine is very essential for the metabolism process as well as neurological function in livestock. Deficiency of this vitamin causes neurological disorders, reduced feed intake, decreased body weight in ruminants and poultry which leads to severe economic losses to the livestock farmers. Many plant-based thiamine sources are available due to their polar nature; they are unstable and possess low bioavailability. Nanoencapsulation technologies, such as nano herbosomes protect vitamins from degradation increase the bioavailability of such phytoconstituents. Thiamine Rich herbosome formulation was developed using thiamine-rich plant sources such as red rice bran, black rice bran, and spirulina with Phosphatidylcholine as carrier. Acute oral toxicity study was conducted using limit test following OECD 423 guidelines. After 14 days of study, no major changes observed in feed intake, body weight, behaviour and gross pathology. No mortality was observed during the course of study. The prepared herbosome formulation has a LD50 > 2000 mg/kg, classifying it as Category-5 (non-toxic) according to the Globally Harmonized System (GHS) of classification, and safe for further therapeutic evaluation.
