Polymorphism study on Exon 2 and 3 of MHC class II DRA gene in Deoni Cattle

Abstract: The present study was undertaken to study polymorphism in MHC class II DRA gene of sixty Deoni cattle maintained at Instructional Livestock Farm Complex, College of Veterinary Science, Rajendranagar A 847 bp genomic region corresponding to the Exon 2 and Exon 3 of MHC class II DRA gene was digested with Taq I and Hae III restriction enzymes for polymorphism. Taq I digestion yielded five fragments of different sizes (768bp, 587bp, 184bp, 112bp, 79bp) in 12 samples and in the remaining 48 samples; it yielded three fragments (587bp, 184bp, 112bp). Hae III digestion has yielded three restriction patterns, 782 & 65bp (in 12 Deoni cattle); 552, 169, & 126bp (in 39 Deoni cattle) and 782, 552, 169, 126 & 65bp (in 9 Deoni cattle). The absence of much variation in BoLA-DRA supports its highly conserved nature and attributed to its recent separation in evolutionary process and/or possibly similar selection pressure to which the ruminants have undergone during the evolutionary process.
