Mast Cell Tumours (MCTs) in dogs: Cytological, histopathological, immunohistochemical, immunocytochemical and molecular diagnosis

Abstract: Mast cell tumours were diagnosed in 177 dogs based on cytological examination of samples from tumour suspected masses. The highest incidence was recorded in non-descript, Labrador and Pug breeds, in male dogs, in dogs of age group 5-10 years and in limbs in breed, sex, age and in location wise analysis. Cytologic examination revealed uniform sized round cells exhibiting anisocytosis and containing varying amounts of metachromatic granules in cytoplasm. Histopathological examination revealed round neoplastic cells containing granules and arranged in linear fashion and sheets. Immunohistochemical investigation revealed positive staining with Vimentin and c kit markers. PCR showed mutation in c kit gene (9.09%). Real time qRT PCR of c kit gene showed significant changes in gene expression of c kit in MCTs.
